The most well known would still be satisfaction. Example, when watching the superbowl a few years ago with my dad and his friends, all between 75 and 80, When the Stones did Satisfaction in their 3 song set, they all aknowledged that was the one song they knew from the Stones. As Stones fans, we can make many arguments for this, but the most universally known, which I guess to me would be the same as the song that best definds them, I would have to say Satisfaction, and no, its not nearly my favorite.
I think satisfaction is even more so. No other song is so recognizalbe and closely associated with any other artist IMO. Maybe Dylan and LARS has that too.
now that I think about it, I know its a compleley different genre but the Beastie Boys "Fight For Your Right To Party" has that quality too. THey have large body of work, but that song is the one that stands out.

But if the signature song is the song that most describes what the band is about or gives an idea of the sound they defined, then either Brown Sugar or JJF have to win the day.

in that case I might go with All Down The Line including live versions. Its this certian combination of rock and soul that only the stones do.

Well, yeah, that's about what I meant ;-). Just imagine someone who never heard the Stones asks you to let them hear 1 song that gives them the best idea what they sound like.
So that doesn't necessarily have to be your favourite song or their most famous one (Satisfaction, without a doubt).
...hey 5string how's it going? (again, what kind of mother names her child 5string?).......my vote goes for Satisfaction. It is the first Stones song I remember hearing. I guess I was 8 years old when I first heard it (in 1966?) when my school buddy's Mom was dancing in their kitchen to Satisfaction on the radio......and she was hot!!! I'll never forget that moment. Charles, my buddy, wanted to go outside to play. I remember saying, "I'll be with you in a minute. I'm gonna hang out in the kitchen for a few more minutes."
